PLEASE HELP!! battery not charging

hey guys
when i take my + battery cable off my eng dies . my car is a 68 firebird with ext regulator. i have not ruled out that it is the alt or the regulator but they are newer parts so i dont realy think it is them.
i think it is in the wiring some were . what do i need to do ? an does the car need to be runing while i check them ? any help would be great.
